【背景】限制性内切酶Mlu I是一种常用的工具酶,在分子生物学领域发挥着重要的作用,其三维结构尚未被解析。【目的】在大肠杆菌中克隆表达、纯化重组Mlu I蛋白及其硒代蛋白,并进行结晶条件的研究。【方法】构建重组表达载体pET28b-Mlu I,在大肠杆菌BL21(DE3)pLysS中诱导表达,利用亲和层析和凝胶过滤层析纯化重组Mlu I蛋白和硒代Mlu I蛋白。对蛋白进行质谱检测、圆二色谱检测以及酶活检测,利用坐滴法进行结晶条件的筛选。【结果】构建了重组表达载体pET28b-Mlu I并纯化获得达到结晶纯度的蛋白,通过质谱检测确定硒代Mlu I蛋白中的8个甲硫氨酸全部被取代,结合酶活测试及圆二色谱检测确定了硒代对Mlu I蛋白的活性、结构无明显影响。采用坐滴法进行初步的晶体生长研究,重组蛋白目前已在1种条件下获得针状晶体并进行初步衍射,获得分辨率在0.32 nm左右的衍射数据。【结论】Mlu I蛋白及硒代Mlu I蛋白纯化体系的构建和结晶条件的研究,可为下一步解析Mlu I三维结构、作用机制的探讨及定向改造奠定基础。  相似文献

Carotenoids are isoprenoid compounds synthesized by all photosynthetic and some non-photosynthetic organisms. They are essential for photosynthesis and contribute to many other aspects of a plant's life. The oxidative breakdown of carotenoids gives rise to the formation of a diverse family of essential metabolites called apocarotenoids. This metabolic process either takes place spontaneously through reactive oxygen species or is catalyzed by enzymes generally belonging to the CAROTENOID CLEAVAGE DIOXYGENASE family. Apocarotenoids include the phytohormones abscisic acid and strigolactones (SLs), signaling molecules and growth regulators. Abscisic acid and SLs are vital in regulating plant growth, development and stress response. SLs are also an essential component in plants’ rhizospheric communication with symbionts and parasites. Other apocarotenoid small molecules, such as blumenols, mycorradicins, zaxinone, anchorene, β-cyclocitral, β-cyclogeranic acid, β-ionone and loliolide, are involved in plant growth and development, and/or contribute to different processes, including arbuscular mycorrhiza symbiosis, abiotic stress response, plant–plant and plant–herbivore interactions and plastid retrograde signaling. There are also indications for the presence of structurally unidentified linear cis-carotene-derived apocarotenoids, which are presumed to modulate plastid biogenesis and leaf morphology, among other developmental processes. Here, we provide an overview on the biology of old, recently discovered and supposed plant apocarotenoid signaling molecules, describing their biosynthesis, developmental and physiological functions, and role as a messenger in plant communication.  相似文献

ODB基因在植物同源重组依赖性的DNA双键断裂修复过程中起重要作用,对植物诱变育种具有潜在的应用价值。克隆烟草NtODB基因并分析其表达特征为丰富ODB基因在同源重组DNA修复过程中的作用提供证据。为得到烟草NtODB基因序列,采用电子克隆技术获得该基因cDNA序列并克隆验证。进一步使用生物信息学方法分析该基因表达特征,对预测蛋白的理化性质、信号肽、高级结构等进行预测。生物信息学分析结果表明,NtODB基因开放阅读框包含579个碱基,蛋白含192个氨基酸残基,NtODB蛋白具有碱性和亲水性,主要定位于细胞质内;实时荧光定量PCR检测结果显示NtODB基因在不同组织中呈现组成型表达特征;亚细胞定位检测提示NtODB主要表达于细胞膜和叶绿体。NtODB基因的克隆与表达分析及其蛋白高级结构和理化性质的预测,可为进一步丰富ODB基因在同源重组依赖的DNA修复系统中的作用机制提供证据。  相似文献

Small fish are highly associated with submerged macrophytes but may potentially hamper their growth due to nutrient excretion that stimulate growth of phytoplankton and periphyton growth. We conducted a mesocosm experiment to elucidate the effects of the small omnivore Chinese bitterling Acheilognathus macropterus on the growth of phytoplankton, periphyton and the submerged macrophyte Vallisneria denseserrulata. The treatments were fishless as well as low (LF) and high (HF) fish density. We found that the concentrations of nutrients and the phytoplankton biomass increased substantially in both fish treatments, leading to a significantly higher light attenuation compared with the control. Moreover, bitterling substantially enhanced the biomass of periphyton on plant leaves. Consequently, the relative growth rate (RGR) of V. denseserrulata was significantly suppressed in HF, while RGR in the LF treatment did not differ significantly from the controls. However, the bitterling also stimulated the ramet production of V. denseserrulata, significantly. Our results indicate that Chinese bitterling reduce the RGR of V. denseserrulata under high fish density condition. Therefore, the density of Chinese bitterling should be kept low in order to reduce the negative effects of the fish on the RGR of submerged macrophytes (e.g. V. denseserrulata), when restoring lakes by plant transplantation.

Ptk2b has been found playing critical roles in oocyte maturation and subsequent fertilization in vitro. But what is the exact in vivo function in reproduction still elusive. Here, by constructing Ptk2b mutant mice, we found Ptk2b was not essential for mice fertility, unexpectedly, contrary to previously reported in vitro findings, we found Ptk2b ablation significantly improved female fecundity. Follicle counting indicated that the number of primordial follicles and growing follicles in matured mice was significantly increased in the absence of Ptk2b, whereas the primordial follicle formation showed no defects. We also found this regulation was in an autophosphorylation independent pathway, as autophosphorylation site mutant mice (PTK2BY402F) show no phenotype in female fertility. Further biochemistry studies revealed that Ptk2b ablation promotes folliculogenesis via Erk pathway mediate follicle survival. Together, we found a novel biological function of Ptk2b in folliculogenesis, which could be potentially used as a therapeutic target for corresponding infertility.  相似文献

人乳头瘤病毒6型(Human papillomavirus type 6,HPV6)是引起生殖器疣与复发性喉乳头瘤的主要病原体之一.为明确2019年济南市1例尖锐湿疣患者的病毒基因组序列特征,本研究提取其尖锐湿疣组织标本总DNA,分两段进行HPV6全基因组PCR扩增和步移法Sanger测序,将拼接后的序列与全球36条不同来源的HPV6全基因组序列进行对比分析.结果 显示,1013/19/JN/CHN/HPV6株(以下简称1013/HPV6)基因组全长8031bp,属于变异谱系B1,与全球不同地区HPV6分离株序列的同源性为98.4%~99.9%.1013/HPV6株与B1亚谱系参考株AF092932全基因组序列相比具有19个核苷酸变异位点,分布在7个开放阅读框架(Open Reading Frames,ORFs)和非编码区内.本研究首次分析了分离自中国大陆的HPV6全基因序列特征,研究结果为HPV分子进化的进一步研究和分型诊断试剂的优化提供了数据.  相似文献

外泌体在细胞生理病理活动过程中起着重要的调控作用,研究外泌体的行为特性对于揭示生命活动及疾病发生发展的内在机理具有重要的基础意义.然而由于缺乏合适的观测手段及方法,目前对于活体状态下外泌体结构及特性的认知仍然很不足.原子力显微镜(AFM)的发明为研究溶液环境下天然状态生物样本提供了强大的技术工具,已成为生物学重要研究手段.本文利用AFM对单个活体状态外泌体的纳米结构及机械特性进行了研究.通过多聚赖氨酸静电吸附作用将从淋巴瘤患者骨髓中分离的外泌体吸附至基底,在溶液环境下实现了对单个活体状态外泌体的高质量AFM形貌成像并通过与空气中成像结果进行对比揭示了空气干燥处理对外泌体形貌的影响.在此基础上,分别利用AFM压痕试验和多参数成像技术实现了对单个活体状态外泌体机械特性的定量测量和可视化表征.最后基于所建立的方法技术揭示了化学处理后外泌体结构和机械特性的动态变化.研究结果为研究纳米尺度下活体状态外泌体的结构及特性,以更好理解天然状态外泌体的生理行为提供了新的方法和思路,对于外泌体研究具有潜在积极的意义.  相似文献

Conservation tillage in conjunction with straw mulching is a sustainable agricultural approach. However, straw mulching reduces the soil temperature, inhibits early maize growth and reduces grain yield in cold regions. To address this problem, we investigated the effects of inoculation of plant growth-promoting rhizobacteria (PGPR) on maize growth and rhizosphere microbial communities under conservation tillage in Northeast China. The PGPR strains Sinorhizobium sp. A15, Bacillus sp. A28, Sphingomonas sp. A55 and Enterobacter sp. P24 were isolated from the maize rhizosphere in the same area and inoculated separately. Inoculation of these strains significantly enhanced maize growth, and the strains A15, A28 and A55 significantly increased grain yield by as much as 22%–29%. Real-time quantitative PCR and high-throughput sequencing showed that separate inoculation with the four strains increased the abundance and species richness of bacteria in the maize rhizosphere. Notably, the relative abundance of Acidobacteria_Subgroup_6, Chloroflexi_KD4-96, and Verrucomicrobiae at the class level and Mucilaginibacter at the genus level were positively correlated with maize biomass and yield. Inoculation with PGPR shows potential for improvement of maize production under conservation tillage in cold regions by regulating the rhizosphere bacterial community structure and by direct stimulation of plant growth.  相似文献
